Biography
Professor Margaret Harnett's research career has been concerned with the study of immune system cell signalling.
For some 30 years, Professor Harnett has been working on the signalling mechanism underpinning development of the immune response in order to identify novel drug targets and immunomodulatory therapies to combat infection and inflammatory disease.
Margaret spent the opening years of her research career as a biochemist investigating pathogen (bacteria and helminth)-specific metabolic pathways in order to identify novel drug targets.
Follwing a PhD at the University of Glasgow, she then worked on innate cell, particularly neutrophil, responses and their dysfunction in diseases at University College London before switching to adaptive B and T cell Biology at the National Institute for Medicine, London.
Since returning to the University of Glasgow, her major focus has been to identify the targets of the anti-inflammatory actions of ES-62 in order to identify biomarkers of inflammatory disease and potential sites of intervention in Asthma, Rheumatoid arthritis, Systemic Lupus Erythematosus and Cardiovascular Disease.
Her major contributions to the field include:
- Role of G-proteins in the immune system: for example, the identification of a novel site of G-protein-regulated exocytosis in neutrophil degranulation (Ge) that was the first step in our understanding of how GTPases regulate endocytic/secretory pathways.
- The discovery that a defect in the respiratory burst of autosomal recessive patients with chronic granulomatous disease reflected their failure to phosphorylate a key component of the NADPH oxidase complex and this contributed to their immunodeficiency.
- Discovery that differential lipid and kinase signalling provides functional checkpoints in B cell biology, potentially impacting on autoimmunity: for example, differential ERK kinetic signatures in immature B cells dictates either cell survival or negative selection (apoptosis), the latter via coupling to phospholipase A2 in a novel caspase-independent pathway.
- Demonstration that remodelling of pulmonary, but not systemic, arteries under conditions of chronic hypoxia reflects enhanced p38 MAPK activation, providing a molecular mechanism underpinning pulmonary hypertension
- Translation of the analysis of intracellular signalling mechanisms from the test-tube to the in situ physiological microenvironment of the immune response in animal models, exploiting Laser Scanning Cytometry.
- Dissection of the molecular mechanisms underpinning evasion of the immune system by pathogens: in particular, the discovery that the parasitic worm product ES-62 subverts TLR4 signalling to induce hyporesponsiveness by downregulating MyD88 and this has identified novel targets for therapeutic intervention in inflammatory disease. This research has additionally led to the generation of ES-62-mimicking compounds that can potentially be developed as novel anti-inflammatory drugs.
Research interests
The major focus of Professor Harnett's laboratory is to understand the differential signalling mechanisms underlying functional maturation of the immune response and its evasion by pathogens in order to identify novel targets for therapeutic intervention in inflammatory disease and to promote healthy ageing.
A particular interest has been to translate the analysis of intracellular signalling mechanisms from the test tube to the in situ physiological environment of the immune response in animal models.
